Thoroughbred horse. The thoracic aorta was removed 1 hour post-mortem on 9/18/85. The culture was initiated using cells released from the lumen of the vessel by collagenase digestion. Cell morphology is polygonal. The cells were grown on gelatin-coated flasks in medium supplemented with horse plasma-derived serum and heparin (0.1mg/ml). The culture was frozen in medium with fetal bovine serum, ECGS (0.02 mg/ml) and heparin (0.05 mg/ml). The culture tested positive forthe endothelial cell-specific von Willebrand factor and negative for alpha-smooth muscle actin (immunofluorescence assays). For lifespan determination this cell line was cultured in Ham's F12 medium and MCDB104 medium in a 1/1 mixture with 10% inactivated fetal bovine serum and supplements (0.04-0.06mg/ml crude ECGF and 0.05mg/ml heparin). Culture medium: AMMC 20U with supplements. Other cultures from this horse are AG08419, AG08420, and AG08422. This cell line will only be shipped frozen. |
